Check the IIS & NTFS permission on the directory. Make sure that the IIS users have access to the folders used by Virtual Server, they're normally called IUSR_machinename and IWAm_machinename, see if that makes a difference.

Unfortunately, I don't know, off hand, what these directories are. I believe one is created in the Documents folder for the user account.
